Overview

Docetaxel + gefitinib is a combination of two small molecule anticancer agents used in clinical trials for the treatment of various cancers, including non-small cell lung cancer (NSCLC) and metastatic breast cancer (MBC). Docetaxel is a taxane that stabilizes microtubules, thereby inhibiting cell division and inducing apoptosis. Gefitinib is an orally active, reversible inhibitor of the epidermal growth factor receptor (EGFR) tyrosine kinase, blocking downstream signaling pathways involved in tumor proliferation and survival. The rationale for combining these drugs lies in their complementary mechanisms—docetaxel targets mitosis while gefitinib inhibits growth signaling—potentially enhancing antitumor efficacy. Clinical studies have shown that this regimen can be active and generally well tolerated, though toxicity profiles may differ from monotherapy regimens[1][2][3][4][5].
