**Docetaxel + icrucumab** is an investigational combination therapy comprising **docetaxel**, a taxane-class cytotoxic chemotherapy agent that inhibits microtubule depolymerization, and **icrucumab**, a recombinant human IgG1 monoclonal antibody targeting vascular endothelial growth factor receptor 1 (VEGFR-1). Docetaxel disrupts mitosis by stabilizing microtubules, leading to cell cycle arrest and apoptosis. Icrucumab inhibits VEGFR-1 activation by blocking ligand binding, thus impairing angiogenesis and tumor growth. This combination was evaluated in phase II trials for the treatment of locally advanced or metastatic urothelial carcinoma after failure of platinum-based chemotherapy, but did not show a significant benefit in progression-free survival compared to docetaxel monotherapy[1][2][3][4].
