Overview

Docetaxel + ifosfamide is a combination chemotherapy regimen consisting of two cytotoxic agents used primarily in the treatment of advanced or refractory solid tumors, including non-small cell lung cancer, urothelial cancer, and soft tissue sarcoma. Docetaxel is an antimicrotubule agent (taxane class) that inhibits microtubule depolymerization, thereby disrupting mitosis and inducing apoptosis in rapidly dividing cells. Ifosfamide is an alkylating agent (oxazaphosphorine class) that crosslinks DNA strands, leading to inhibition of DNA replication and cell death. The combination has been studied in phase I and II trials for patients with advanced cancers who have failed prior therapies. The regimen has demonstrated antitumor activity with manageable toxicity profiles; main toxicities include neutropenia, anemia, thrombocytopenia, gastrointestinal symptoms (nausea/vomiting), mucositis, neuropathy, hypersensitivity reactions, and edema[1][2][4].
