This is a combination therapy consisting of docetaxel, a microtubule inhibitor (taxane class), and imexon, an aziridine-based small molecule pro-oxidant. Docetaxel disrupts microtubule function leading to cell cycle arrest and apoptosis in cancer cells. Imexon induces oxidative stress by increasing reactive oxygen species (ROS) within tumor cells, resulting in mitochondrial dysfunction and activation of the intrinsic apoptotic pathway via caspases 3 and 9. The combination has shown synergistic antitumor activity in preclinical models and early-phase clinical trials for advanced solid tumors such as breast cancer, prostate cancer, and non-small cell lung cancer (NSCLC). Clinical studies have established the maximum tolerated dose for this regimen and demonstrated preliminary evidence of efficacy[1][3][5].
