Overview

Docetaxel + irinotecan is a combination chemotherapy regimen evaluated by the National Cancer Center, Korea, for the treatment of recurrent or refractory bone and soft tissue sarcomas, including Ewing sarcoma. Docetaxel is a small molecule taxane that binds to and stabilizes microtubules, preventing their depolymerization and leading to cell cycle arrest at the G2/M phase. Irinotecan is a small molecule prodrug that is converted to its active metabolite, SN-38, which inhibits DNA topoisomerase I, causing double-strand DNA breaks and subsequent apoptosis. This combination therapy was specifically investigated in a Phase 2 clinical trial (NCT01380275) targeting patients under 50 years of age with advanced sarcomas.
