Docetaxel + letrozole is a combination therapy consisting of two small molecule drugs—docetaxel, a taxoid anticancer agent, and letrozole, a non-steroidal aromatase inhibitor. Docetaxel stabilizes microtubules by binding to the β-subunit of tubulin, preventing their depolymerization and thereby disrupting cell division and inducing cancer cell apoptosis[1]. Letrozole competitively inhibits the CYP19A1 (aromatase) enzyme, preventing the conversion of androgens into estrogens and thereby suppressing estrogen-dependent tumor growth[2]. The combination is sometimes used in the treatment of estrogen receptor-positive metastatic breast cancer, leveraging complementary mechanisms where letrozole suppresses tumor-promoting estrogen while docetaxel inhibits cell proliferation and survival pathways.
