Docetaxel + lonafarnib is a combination of two small molecule drugs investigated primarily for the treatment of solid tumors, including hormone-refractory prostate cancer and other refractory solid tumors. Docetaxel is a taxoid antineoplastic agent that promotes microtubule assembly and stabilizes microtubules by preventing their depolymerization, thereby inhibiting cell division and inducing apoptosis in cancer cells. Lonafarnib is a potent farnesyl transferase inhibitor (FTI) that blocks the farnesylation of proteins such as progerin and Ras, interfering with oncogenic signaling pathways. Preclinical studies have shown that this combination has synergistic or additive anti-tumor effects, enhancing inhibition of cell proliferation and apoptosis compared to either agent alone. The combination has been evaluated in clinical trials for refractory solid tumors to assess safety, tolerability, biological activity, and preliminary efficacy[2][3].
