The combination of docetaxel, nedaplatin, and fluorouracil (often abbreviated as the DNF regimen) is a multi-agent chemotherapy protocol primarily used for the treatment of advanced or recurrent esophageal cancer and has also been studied in locoregionally advanced nasopharyngeal carcinoma. - **Docetaxel** is a taxane-class antineoplastic agent that promotes microtubule assembly while inhibiting their disassembly, thereby disrupting mitosis and leading to cell death. - **Nedaplatin** is a platinum-based chemotherapeutic similar to cisplatin but with reduced nephrotoxicity; it forms DNA crosslinks that inhibit DNA replication and transcription. - **Fluorouracil (5-FU)** is an antimetabolite that inhibits thymidylate synthase, blocking DNA synthesis in rapidly dividing cells. This triplet regimen has demonstrated efficacy as neoadjuvant therapy before surgery or as first-line/second-line therapy for unresectable or recurrent esophageal squamous cell carcinoma[1][2][3][4][8]. It has also shown promising results in induction chemotherapy for nasopharyngeal carcinoma[5]. The combination leverages synergistic cytotoxic effects from each component’s distinct mechanism of action.
