This is a combination therapy consisting of three agents: **Docetaxel** is a taxane-class small molecule chemotherapeutic that stabilizes microtubules, thereby inhibiting cell division and inducing apoptosis in cancer cells. It is widely used for the treatment of various cancers, including breast, lung, and prostate cancer. **REOLYSIN** (pelareorep) is an oncolytic virus therapy based on a proprietary variant of reovirus. It preferentially infects and replicates inside cancer cells with activated Ras pathways, leading to direct tumor cell lysis and stimulating an anti-tumor immune response by releasing tumor antigens[6][4][8]. **Prednisone** is a synthetic glucocorticoid corticosteroid that exerts anti-inflammatory and immunosuppressive effects. In prostate cancer regimens, it helps reduce inflammation and may slow tumor growth by decreasing testosterone production[5]. The combination has been studied primarily in metastatic castration-resistant prostate cancer as well as other advanced solid tumors[2][4]. Docetaxel plus prednisone was FDA-approved for metastatic hormone-refractory prostate cancer based on improved survival outcomes compared to mitoxantrone plus prednisone[1]. The addition of REOLYSIN aims to enhance anti-tumor efficacy through synergistic mechanisms involving both direct cytotoxicity and immune activation.
