The TTC regimen is a combination chemotherapy treatment consisting of docetaxel (Taxotere), temozolomide (Temodar), and cisplatin, which was evaluated in a Phase I clinical trial at the M.D. Anderson Cancer Center for patients with metastatic melanoma. Docetaxel is a taxane that promotes microtubule assembly and inhibits disassembly, leading to cell cycle arrest in the G2/M phase. Temozolomide is an oral alkylating agent that undergoes rapid chemical conversion to the active monomethyl triazene (MTIC), which methylates DNA at the O6 and N7 positions of guanine. Cisplatin is a platinum-based antineoplastic agent that forms covalent bonds with DNA bases, creating intra-strand and inter-strand cross-links that interfere with DNA replication and transcription. This triple-drug combination was investigated to determine the maximum tolerated dose and assess its efficacy in treating advanced melanoma, a disease typically resistant to single-agent chemotherapy.
