Drug intelligence / Profile preview
PR-1 is a liposomal nanoparticle formulation of the cytotoxic agent docetaxel, developed by Puripharm. Docetaxel is a taxane that promotes the assembly of tubulin into stable microtubules and inhibits their disassembly, which leads to a marked decrease in free tubulin. This stabilization interferes with the normal dynamic reorganization of the microtubule network that is essential for vital interphase and mitotic cellular functions, ultimately inducing cell cycle arrest and apoptosis. The liposomal formulation is intended to enhance the therapeutic index of docetaxel by improving its solubility, prolonging circulation time, and increasing tumor-specific delivery via the enhanced permeability and retention (EPR) effect, while potentially mitigating side effects associated with traditional surfactants like polysorbate 80.
