Doce NC (BMS-582483) is a specialized nanocrystal formulation of the antineoplastic agent docetaxel, developed by Bristol-Myers Squibb. This formulation utilizes nanocrystal technology to create a submicron suspension of docetaxel, which improves the drug's solubility and eliminates the need for surfactants like Polysorbate 80 (Tween 80) and ethanol, which are required in the standard docetaxel formulation (Taxotere) and are often responsible for severe hypersensitivity reactions. By reducing the need for these excipients, Doce NC aims to improve the safety and tolerability profile of docetaxel therapy. Mechanistically, Doce NC functions as a microtubule stabilizer; it binds to the beta-subunit of tubulin, promoting the assembly of microtubules and preventing their disassembly. This stabilization disrupts the dynamic equilibrium of the microtubule network, leading to mitotic spindle dysfunction, cell cycle arrest at the G2/M phase, and ultimately, programmed cell death. Doce NC has been primarily developed and approved in certain international markets, such as Thailand, for the treatment of advanced non-small cell lung cancer (NSCLC), breast cancer, and other taxane-responsive malignancies.
