Overview

Docetaxel thermosensitive liposome is a heat-activated drug delivery system developed by IMUNON (formerly Celsion Corporation). It consists of the potent taxane chemotherapy agent docetaxel encapsulated within specialized temperature-sensitive liposomes. These liposomes are designed to remain stable at normal body temperature (37°C) but undergo a rapid phase transition when exposed to mild hyperthermia (typically 40-42°C), triggering the localized release of the drug. This targeted approach aims to achieve significantly higher concentrations of docetaxel within the tumor microenvironment—potentially more than double that of conventional docetaxel—while reducing systemic toxicity. Once released, docetaxel functions as a microtubule inhibitor, binding to tubulin to stabilize microtubules and prevent their disassembly, which leads to cell cycle arrest and apoptosis in rapidly dividing cancer cells.
