**Docosahexaenoic acid + doxorubicin** is a combination of docosahexaenoic acid (DHA), a long-chain omega-3 polyunsaturated fatty acid, and doxorubicin, an anthracycline chemotherapeutic agent. This combination is being investigated primarily for the treatment of **breast cancer**, especially in chemoresistant or recurrent tumors. Preclinical and pilot clinical studies indicate that DHA can enhance the cytotoxicity of doxorubicin, delay or inhibit tumor recurrence, and potentially reduce systemic toxicity when co-encapsulated with doxorubicin in specialized formulations such as nanoemulsions or nanostructured lipid carriers. The synergistic antitumor activity is mediated in part by modulation of cellular pathways including attenuation of TLR4 activation, induction of apoptosis, inhibition of cancer cell proliferation, increased intracellular accumulation of doxorubicin, cell cycle arrest at the G2/M phase, reduction of multidrug resistance proteins, and a decrease in pro-inflammatory and angiogenic factors within the tumor microenvironment[1][2][3][6]. The combination has also been examined for its cardioprotective potential, as DHA may attenuate doxorubicin-induced cardiotoxicity[4].
