Dodekin (IL12-L19L19) is a human antibody-cytokine fusion protein composed of interleukin-12 (IL-12) fused to the L19 antibody. The L19 antibody targets the extra-domain B of fibronectin (EDB-FN), which is abundantly expressed in the extracellular matrix of tumor vasculature. By targeting EDB-FN, IL12-L19L19 delivers IL-12 directly to the tumor microenvironment, aiming to enhance local immune activation while minimizing systemic toxicity. The mechanism of action involves stimulation of the interleukin-12 receptor on immune cells, leading to increased production of interferon-gamma and activation of cytotoxic T lymphocytes and natural killer cells for anti-tumor activity. Dodekin is being developed primarily for advanced or metastatic solid tumors after prior immune checkpoint blockade therapy[1][4][5][7].
