Drug intelligence / Profile preview
Dodoviscin H is a natural isoprenylated flavonol derivative isolated from the aerial parts of *Dodonaea viscosa*. It is under preclinical development by Peking University and Peking University First Hospital as a potential targeted therapy for membranous nephropathy (MN). Dodoviscin H acts by promoting the selective degradation of the podocyte autoantigen phospholipase A2 receptor (PLA2R). Mechanistically, chemoproteomics studies have shown that Dodoviscin H binds to Rabaptin-5 (RABEP1) and Hsc70 (HSPA8), synergistically driving PLA2R degradation through the Rabaptin-5-mediated endosome-lysosome pathway and the Hsc70-mediated ubiquitination-proteasome pathway. In preclinical models, Dodoviscin H has demonstrated the ability to protect human podocytes from membranous nephropathy patient plasma and selectively reduce kidney PLA2R levels in transgenic mice.
