Dolastatin 15 is a **linear depsipeptide** derived from the marine mollusc Dolabella auricularia. It is a **potent antimitotic and cytostatic agent** that exerts its effects primarily by interfering with microtubule assembly, resulting in G2/M cell cycle arrest and subsequent apoptosis in various cancer cell types[1][2][4]. Its mechanism of action, while structurally related to dolastatin 10, is characterized by a weaker in vitro interaction with tubulin but robust antineoplastic activity in cell lines and animal models, including notable effects in myeloma, lymphoma, leukemia, breast, lung, ovarian, and colon cancer cells[1][2][4]. Dolastatin 15 induces apoptosis through both mitochondrial pathways and Fas (CD95)-mediated pathways[2]. It is under investigation as an antineoplastic agent; however, it is less potent than dolastatin 10 in direct comparisons[4]. Derivatives of dolastatin 15 have served as payloads in antibody-drug conjugate (ADC) research[3].
