Drug intelligence / Profile preview
Dolasynthen DAR-2 ADC refers to a class of antibody-drug conjugates (ADCs) developed using Mersana Therapeutics' proprietary Dolasynthen platform, characterized by a drug-to-antibody ratio (DAR) of 2. The Dolasynthen platform utilizes a fully synthetic, biodegradable scaffold to enable precise, site-specific conjugation of auristatin-based payloads (such as auristatin hydroxypropylamide, or AF-HPA) to targeting antibodies. In preclinical studies, Dolasynthen DAR-2 ADCs (such as those targeting B7-H4 or HER2) have been evaluated as comparators alongside higher DAR variants (e.g., DAR-6 and DAR-12) to optimize the therapeutic index, pharmacokinetics, and efficacy of clinical candidates like XMT-1660.
