DOM-CAP-1 is a DNA fusion vaccine designed to target carcinoembryonic antigen (CEA)-expressing cancers. It consists of the HLA-A*0201–binding peptide CAP-1, derived from CEA, linked to an immunostimulatory domain (DOM) from fragment C of tetanus toxin. This vaccine aims to stimulate CD8+ T-cell immune responses against the CAP-1 peptide by exploiting the nontolerized CD4+ T-cell repertoire for tetanus, thereby overcoming peripheral tolerance to tumor-associated antigens. It was developed by Cancer Research UK and produced by the NHS Blood and Transplant, Clinical Biotechnology Centre, University of Bristol.
