Domagrozumab is a humanized monoclonal antibody developed by Pfizer for the treatment of Duchenne muscular dystrophy (DMD) and investigated for limb girdle muscular dystrophies. It acts as an anti-myostatin antibody, binding to and inhibiting myostatin (Growth/differentiation factor 8 (GDF-8)), a protein that negatively regulates skeletal muscle growth. By blocking myostatin activity, domagrozumab was intended to increase muscle mass and strength in patients with muscle-wasting diseases such as DMD. The drug was administered intravenously and underwent clinical trials, including Phase 1 and Phase 2 studies in both healthy volunteers and pediatric DMD patients. Despite initial promise, development was terminated after clinical trials failed to demonstrate significant efficacy[1][3][4][5][7][8].
