Overview

Dominant-negative Mastermind-like 1 (DN-MAML1) viral vector is a gene therapy tool designed to inhibit the Notch signaling pathway. Mastermind-like 1 (MAML1) is a critical co-activator that binds to the Notch intracellular domain (NICD) and CSL (CBF1/RBP-Jκ) to form a transcriptional activation complex. The dominant-negative form of MAML1 typically consists of the N-terminal Notch-binding domain but lacks the transactivation domains; it competitively binds to the NICD/CSL complex, thereby preventing the recruitment of other co-activators and effectively blocking Notch-mediated gene transcription. In oncology research, particularly in melanoma, this vector is used to study the role of Notch signaling in tumor progression, cell growth, and survival by antagonizing the oncogenic effects of aberrant Notch activation.
