Overview

Domvanalimab is a fully humanized Fc-silent monoclonal antibody targeting the T-cell immunoreceptor with Ig and ITIM domains (TIGIT). It is designed to block the inhibitory effect of TIGIT on immune cells in the tumor microenvironment, thereby "releasing the brakes" on anti-cancer immunity and allowing activation of immune pathways that promote tumor cell death. By being Fc-silent, domvanalimab avoids inducing antibody-dependent cellular cytotoxicity (ADCC), which helps prevent depletion of regulatory T cells and reduces risk of autoimmune toxicities. Domvanalimab is under investigation in combination with other immunotherapies such as zimberelimab (anti-PD-1) and chemotherapy for various cancers including non-small cell lung cancer (NSCLC), hepatobiliary cancers, melanoma, bladder cancer, gastric/gastroesophageal junction cancers[2][4][7].
