A combination regimen consisting of domvanalimab (an Fc-silent monoclonal antibody targeting TIGIT), zimberelimab (a monoclonal antibody targeting PD-1), and a platinum-based chemotherapy doublet (commonly cisplatin or carboplatin combined with a third-generation agent such as pemetrexed, paclitaxel, gemcitabine, vinorelbine, or docetaxel). Domvanalimab and zimberelimab co-inhibit TIGIT and PD-1 pathways, enhancing immune activation against tumors. The platinum-based doublet delivers cytotoxic chemotherapy, standard for first-line treatment in advanced non-small cell lung cancer (NSCLC). This combination is being evaluated as a first-line treatment for metastatic NSCLC (including both squamous and nonsquamous histologies) and for advanced gastrointestinal cancers in late-phase clinical trials[1][2][4][5][6].
