Overview

A combination therapy consisting of **donafenib**, **cytarabine**, and **daunorubicin** used investigationally in the treatment of *relapsed acute myeloid leukemia (AML)*. **Donafenib** is an oral multikinase inhibitor targeting Raf kinase and various receptor tyrosine kinases. **Cytarabine** is an antimetabolite interfering with DNA synthesis, especially during the S-phase of the cell cycle. **Daunorubicin** is an anthracycline chemotherapeutic agent that intercalates into DNA, inhibits topoisomerase II, and generates free radicals leading to DNA damage. The rationale for combining these agents is to capitalize on the synergistic anticancer activities to potentially improve outcomes in relapsed AML patients[3].
