Donafenib + PD-1 is a combination therapeutic regimen primarily investigated for the treatment of unresectable advanced hepatocellular carcinoma (HCC). Donafenib is an orally administered small molecule multi-kinase inhibitor that targets vascular endothelial growth factor receptors (VEGFR), platelet-derived growth factor receptors (PDGFR), and the Raf/MEK/ERK signaling pathway to inhibit tumor angiogenesis and proliferation. The PD-1 inhibitor component is a monoclonal antibody that blocks the programmed death-1 (PD-1) receptor on T cells, preventing its interaction with ligands PD-L1 and PD-L2, thereby restoring anti-tumor immune responses. This combination is often evaluated alongside Transarterial Chemoembolization (TACE) to enhance local and systemic control of HCC by combining anti-angiogenic effects with immune checkpoint blockade.
