Overview

This is a combination therapy consisting of donafenib, sintilimab, and hepatic artery infusion chemotherapy (HAIC). Donafenib is an oral small molecule multikinase inhibitor targeting several kinases involved in tumor growth and angiogenesis. Sintilimab is a monoclonal antibody that acts as an immune checkpoint inhibitor by targeting programmed cell death protein 1 (PD-1), thereby enhancing the immune system's ability to attack cancer cells. HAIC involves the direct infusion of chemotherapeutic agents into the hepatic artery to increase drug concentration at liver tumors while minimizing systemic exposure. This combination has been investigated primarily for advanced or unresectable hepatocellular carcinoma (HCC), particularly in patients with high-risk features such as portal vein tumor thrombus[1][2][4][5].
