donanemab

Donanemab is a humanized monoclonal antibody (IgG1) developed for the treatment of early symptomatic Alzheimer's disease, including mild cognitive impairment and mild dementia due to Alzheimer's. It specifically targets the N-terminal, third amino acid pyroglutamate (N3pG) epitope of amyloid beta present in brain plaques. By binding to this modified form of amyloid beta (pyroglutamate Aβ at position 3, pGlu3-Aβ), donanemab promotes removal of amyloid plaques through microglial-mediated phagocytosis, thereby slowing disease progression.
