**Donanemab + LY3202626** is a combination therapy investigated for Alzheimer's disease. Donanemab is a monoclonal antibody that targets and helps to clear amyloid-beta plaques from the brain, developed and marketed under the brand name Kisunla. LY3202626 is a potent, selective small molecule inhibitor of beta-site amyloid precursor protein cleaving enzyme 1 (BACE1), designed to reduce production of pathogenic amyloid-beta peptides by inhibiting their cleavage from amyloid precursor protein. Combination trials aimed to evaluate efficacy and safety of dual amyloid targeting (removal and production blockade) in Alzheimer's. The arm combining intravenous donanemab with oral LY3202626 was discontinued due to low probability of superior clinical benefit over monotherapy. Both agents were developed by Eli Lilly.
