Donor-derived activated natural killer (NK) cells are an allogeneic adoptive cell therapy developed by Beijing Boren Hospital for the treatment of hematological malignancies, specifically acute myeloid leukemia (AML). The therapy involves isolating NK cells from a healthy donor, followed by ex vivo expansion and activation using cytokines such as interleukin-2 (IL-2) or interleukin-15 (IL-15) to enhance their anti-tumor cytotoxicity. Once infused into the patient, these activated NK cells identify and eliminate leukemic blasts through the recognition of stress-induced ligands via activating receptors (e.g., NKG2D, NKp46) and the subsequent release of cytotoxic granules containing perforin and granzymes. This approach is particularly investigated as a consolidation therapy or a bridge to hematopoietic stem cell transplantation in patients with refractory or relapsed AML.
