Donor-derived CIK (Cytokine-Induced Killer) cells are an adoptive cellular immunotherapy developed by A.O. Ospedale Papa Giovanni XXIII for the treatment of relapsed hematologic malignancies following haploidentical stem cell transplantation. These cells are an Advanced Therapeutic Medicinal Product (ATMP) consisting of ex vivo-expanded immune effector cells derived from a haploidentical donor's peripheral blood or leukapheresis product. CIK cells are a heterogeneous population, primarily characterized by the co-expression of T cell (CD3) and Natural Killer (NK) cell (CD56) markers. They exert potent, non-MHC-restricted cytolytic activity against tumor cells, particularly leukemia, while maintaining a low potential for inducing graft-versus-host disease (GvHD). The therapy is currently being evaluated in Phase I/II clinical trials (e.g., Haplo-CIK) for conditions including acute myeloid leukemia (AML), acute lymphoblastic leukemia (ALL), multiple myeloma, myelofibrosis, and myelodysplastic syndrome (MDS).
