Overview

Donor-derived cytomegalovirus-specific cytotoxic T lymphocytes (CMV-CTL) is an adoptive cellular immunotherapy developed by Shanghai General Hospital for the treatment of cytomegalovirus (CMV) infection in patients following allogeneic hematopoietic stem cell transplantation (HSCT). The therapy is produced by isolating peripheral blood mononuclear cells (PBMCs) from the original stem cell donor and stimulating them with CMV antigen peptides over an 18-21 day culture period to induce and expand a population of T cells specifically reactive to CMV. Once infused into the recipient, these cytotoxic T lymphocytes recognize and eliminate CMV-infected cells, thereby restoring the patient's cellular immunity against the virus. This approach aims to provide a targeted treatment for CMV reactivation or infection, which remains a significant cause of morbidity and mortality in immunocompromised transplant recipients.
