Donor-derived latent membrane protein (LMP)-specific cytotoxic T-lymphocytes (CTLs) are an adoptive cellular immunotherapy developed by New York Medical College for the treatment of Epstein-Barr virus (EBV)-positive Hodgkin lymphoma. This therapy involves the ex vivo expansion of T-cells from an HLA-matched donor that are specifically reactive against EBV-associated antigens, specifically Latent Membrane Protein 1 (LMP1) and Latent Membrane Protein 2 (LMP2). These proteins are frequently expressed in EBV-associated malignancies like Hodgkin lymphoma and serve as targets for the immune system. In the clinical setting, these CTLs are infused following a reduced-intensity allogeneic stem cell transplant to provide a targeted graft-versus-lymphoma effect while minimizing the risk of graft-versus-host disease, aiming to eliminate residual EBV-positive tumor cells.
