Overview

Donor-derived multi-infection specific cytotoxic T lymphocytes (CTLs) is an adoptive cellular immunotherapy developed by the University of Sydney for the prevention and treatment of opportunistic infections in patients undergoing allogeneic hematopoietic stem cell transplantation (HSCT). The therapy involves the ex vivo expansion of donor-derived T cells sensitized against a broad panel of antigens from seven common post-transplant pathogens: Cytomegalovirus (CMV), Adenovirus (Adv), Epstein-Barr virus (EBV), Varicella-zoster virus (VZV), Influenza, BK polyomavirus (BKV), and the fungus Aspergillus. By providing a broad repertoire of pathogen-specific T cells, the treatment aims to bridge the period of profound immunodeficiency post-transplant. In the CYNTAX clinical protocol, these CTLs are administered in combination with early vaccination using inactivated influenza and live-attenuated VZV vaccines to promote long-term immune reconstitution and enhance the persistence of the infused T-cell memory.
