Donor Enriched Activated Natural Killer Cell therapy refers to the adoptive transfer of natural killer (NK) cells that are isolated and enriched from a healthy donor, then activated ex vivo—often using cytokines such as IL-2 or IL-15—prior to infusion into a recipient. This approach is primarily used in the context of allogeneic hematopoietic stem cell transplantation (allo-HSCT) or as an adjunct immunotherapy for hematologic malignancies and some solid tumors. The mechanism of action involves direct cytotoxicity against malignant cells via innate immune recognition, without inducing graft-versus-host disease (GVHD), and enhancement of immune reconstitution post-transplant. These therapies may use various sources for NK cells, including peripheral blood or umbilical cord blood from HLA-matched or haploidentical donors.
