Donor Enriched Activated Natural Killer (DEA-NK) cells are an investigational adoptive cellular immunotherapy developed by the University of Nebraska Medical Center for the treatment of refractory myeloid malignancies. The therapy consists of natural killer (NK) cells obtained from a haploidentical (half-matched) donor, which are then enriched and activated ex vivo using cytokines to enhance their anti-leukemic activity and longevity. These activated NK cells are administered intravenously to patients on day +7 following a haploidentical stem cell transplant (Haplo-SCT) that utilizes post-transplant cyclophosphamide (PTCY). The mechanism of action involves natural killer cell-mediated cytotoxicity, where the infused cells directly target and eliminate residual tumor cells in patients with acute myeloid leukemia (AML), myelodysplastic syndromes (MDS), or myeloproliferative neoplasms (MPN).
