Donor lymphocyte infusion (DLI) is an adoptive cellular immunotherapy used to prevent or treat relapse in patients following allogeneic hematopoietic stem cell transplantation (allo-HSCT). The therapy involves the administration of lymphocytes from the original donor to the recipient to harness the graft-versus-leukemia (GVL) effect, where donor T cells identify and destroy residual malignant cells. The Chinese PLA General Hospital has specifically evaluated prophylactic G-CSF-primed DLI (GP-DLI) in patients with high-risk hematologic malignancies, such as leukemia and lymphoma. This protocol may include the use of decitabine as a conditioning or synergistic agent for patients with specific high-risk genetic mutations (e.g., TP53, TET2, DNMT3a) to enhance the anti-tumor efficacy of the infused lymphocytes.
