Donor mesenchymal stem cells (MSCs) are multipotent stromal cells derived from a healthy human donor. They can differentiate into various cell types including osteoblasts (bone), chondrocytes (cartilage), myocytes (muscle), and adipocytes (fat). Donor MSCs are used in allogeneic transplantation settings—meaning the cells come from a different individual than the recipient. Their primary mechanisms include immunomodulation, anti-inflammatory effects, secretion of growth factors and cytokines to promote tissue repair and regeneration, and homing to sites of injury or inflammation. Donor MSCs do not typically engraft long-term but exert their therapeutic effects through paracrine signaling ("hit-and-run" mechanism). They are being investigated for a wide range of indications such as autoimmune diseases (e.g., graft versus host disease), degenerative conditions (e.g., osteoarthritis), inflammatory disorders, tissue repair after injury or surgery, and as carriers for drug delivery in cancer therapy[1][3][7].
