Donor Natural Killer Cell Therapy is an investigational adoptive cellular immunotherapy developed by the Fred Hutchinson Cancer Center for the treatment of various hematologic malignancies. The therapy consists of a single, purified infusion of natural killer (NK) cells derived from an HLA-haploidentical donor, typically the same donor used for a preceding hematopoietic cell transplantation (HCT). Administered on day +7 following nonmyeloablative HCT, these NK cells are intended to provide a pre-emptive graft-versus-tumor (GVT) effect to eliminate residual cancer cells and prevent disease relapse. Because NK cells do not typically cause graft-versus-host disease (GVHD) in the same manner as T cells, this approach aims to enhance the anti-tumor efficacy of the transplant without increasing the risk of severe immune-mediated complications. It is currently being studied in Phase I/II trials for conditions including acute myeloid leukemia, acute lymphoblastic leukemia, and several types of lymphoma and myeloma.
