Overview

Donor regulatory T lymphocytes are a cell therapy product consisting of regulatory T cells (Tregs) derived from a healthy or compatible human donor. These specialized CD4+ lymphocytes play a critical role in immune regulation by suppressing the activation and proliferation of effector T cells, thereby maintaining immune tolerance and preventing excessive immune responses. In clinical settings, adoptive transfer of donor-derived regulatory T lymphocytes is being investigated primarily to prevent graft-versus-host disease (GVHD) following allogeneic hematopoietic stem cell transplantation and to promote tolerance in solid organ transplantation. The mechanism of action involves immunosuppression through direct cell-cell contact and secretion of inhibitory cytokines such as transforming growth factor beta (TGF-β). Donor regulatory T cell therapies are currently under investigation in early-phase clinical trials for their ability to reduce the need for chronic immunosuppression while minimizing transplant rejection[4][5][6].
