Overview

Donor T-lymphocytes, often administered as a Donor Lymphocyte Infusion (DLI), are a form of adoptive cellular immunotherapy used primarily in the setting of allogeneic hematopoietic stem cell transplantation (HSCT). In the specific clinical protocol developed by the M.D. Anderson Cancer Center, these donor-derived CD3+ T-cells are infused into the patient approximately 8-9 weeks following a reduced-intensity transplant. The therapeutic rationale is to induce a "graft-versus-tumor" (GVT) effect, wherein the donor's immune cells recognize and eliminate residual malignant cells in the recipient that may have survived the initial transplant conditioning. This approach is being investigated to reduce the risk of disease relapse in patients with various hematologic malignancies, including leukemia, myelodysplastic syndrome (MDS), lymphoma, Hodgkin lymphoma, and multiple myeloma.
