Drug intelligence / Profile preview
Doparkin is an oral fixed-dose combination of levodopa, a dopamine precursor, and carbidopa, a peripheral dopa decarboxylase (DDC) inhibitor, developed and marketed by General Pharmaceuticals for the treatment of Parkinson's disease and parkinsonian syndromes. Levodopa crosses the blood-brain barrier and is converted into dopamine in the brain, replenishing depleted striatal dopamine levels to alleviate motor symptoms such as rigidity, bradykinesia, and tremors. Because levodopa is rapidly metabolized in peripheral tissues, carbidopa is co-administered to inhibit peripheral decarboxylation. Since carbidopa does not cross the blood-brain barrier, it increases the bioavailability of levodopa to the central nervous system while reducing peripheral dopaminergic side effects like nausea and vomiting.
