TuHURA Therapeutics is developing a bi-functional, bi-specific platform that links a Delta Opioid Receptor (DOR) small molecule antagonist to an immune-modulating antibody, specifically targeting the V-domain Ig suppressor of T cell activation (VISTA). The lead candidate in this program involves conjugating a potent, highly selective DOR inhibitor to the VISTA-inhibiting monoclonal antibody TBS-2025. The mechanism of action involves antagonizing DOR on tumor-associated myeloid-derived suppressor cells (MDSCs) and tumor-associated macrophages (TAMs) to reduce myeloid-mediated immunosuppression and reverse T-cell dysfunction. By reprogramming these suppressive cells in the tumor microenvironment, the therapy aims to enhance anti-tumor immune responses and overcome acquired resistance to checkpoint inhibitors and other immunotherapies in solid tumors.
