Doramectin is a semisynthetic avermectin and macrocyclic lactone used as a veterinary antiparasitic drug. It is approved for the treatment and control of internal and external parasites—including gastrointestinal roundworms, lungworms, eyeworms, grubs, sucking lice, mange mites, ticks, and other ectoparasites—in cattle and swine[1][2][4][5]. Doramectin acts primarily by binding to glutamate-gated chloride channel receptor in the nervous system of nematodes and arthropods. This increases membrane permeability to chloride ions leading to paralysis and death of the parasite[2][6]. It may also enhance GABA release at presynaptic neurons in parasites[6]. Doramectin is produced by fermentation from Streptomyces avermitilis followed by chemical modification[4][6]. It is available as injectable or topical formulations for animals. While not approved for human use or dairy cattle[5], research has explored its potential anticancer effects via autophagy induction through PI3K/AKT/mTOR pathway inhibition in glioblastoma models[8].
