Overview

Doravirine + emtricitabine + tenofovir alafenamide is a combination antiretroviral therapy used in the treatment and prevention of HIV-1 infection. Doravirine is a non-nucleoside reverse transcriptase inhibitor (NNRTI) that inhibits HIV-1 reverse transcriptase, thereby blocking viral replication. Emtricitabine and tenofovir alafenamide are nucleoside/nucleotide reverse transcriptase inhibitors (NRTIs), which also inhibit HIV-1 reverse transcriptase by causing chain termination during viral DNA synthesis. Tenofovir alafenamide is a prodrug of tenofovir, designed to deliver higher concentrations of the active drug to target cells with reduced systemic exposure, improving safety profiles compared to earlier formulations. This combination must be used as part of a complete antiretroviral regimen for treatment or as pre-exposure prophylaxis (PrEP) for prevention in certain populations[5][3][6].
