Overview

Dordaviprone is an oral, first-in-class, CNS-penetrant small-molecule imipridone developed by Oncoceutics and Chimerix and now associated with Jazz's oncology portfolio following the 2025 acquisition of Chimerix. It is being developed primarily for H3 K27M-mutant diffuse glioma, including diffuse midline glioma, and has also been studied in other solid tumors such as endometrial cancer and in colorectal cancer chemoprevention. Mechanistically, dordaviprone antagonizes dopamine receptor D2 and D3 and agonizes the mitochondrial protease ClpP, triggering the integrated stress response with ATF4 and CHOP induction and promoting TRAIL and DR5-mediated apoptosis. Clinical and translational reports also describe brain penetration, generally favorable tolerability, and activity signals in recurrent H3 K27M-mutant diffuse glioma.
