Doripenem is a broad-spectrum, beta-lactam antibiotic of the carbapenem class. It is used to treat complicated intra-abdominal and urinary tract infections caused by susceptible bacteria. Doripenem acts by inhibiting bacterial cell wall synthesis through binding to multiple essential penicillin-binding proteins (PBPs), leading to cell death. It is stable against most beta-lactamases, including extended-spectrum types, but can be inactivated by carbapenemases. Doripenem was developed and marketed under the brand name Doribax, initially launched by Shionogi in Japan and later marketed outside Japan by Johnson & Johnson companies such as Janssen. The drug was approved in 2007 for intravenous use but has since been discontinued in several markets due to safety concerns, particularly an increased risk of death when used for ventilator-associated pneumonia[3][5][7].
