DoriVac is a **DNA origami-based vaccine platform** designed to precisely arrange adjuvants and antigens on nanoparticles, with the aim to enhance antigen-specific immune responses and enable personalized immunotherapy[5][6]. This technology uses nanoscale spatial arrangement of DNA and biomolecules to optimize activation of both the humoral (antibody-mediated) and cellular (T cell-mediated) arms of the immune system. DoriVac nanoparticles can be loaded with disease-specific antigens—such as peptides or intact proteins—and immunostimulatory CpG oligonucleotide adjuvants at controlled intervals, improving uptake and activation of dendritic cells and other antigen-presenting cells, leading to robust CD4+ and CD8+ T cell responses[3][4][1][2]. It has shown promise in preclinical models for **infectious diseases** (SARS-CoV-2, HIV, Ebola) and multiple **cancers** (melanoma, lymphoma, colon cancer, lung cancer, neuroblastoma), with the ability to synergize with immune checkpoint inhibitors to further suppress tumor growth[3][4][5][6][7]. The platform was developed and studied by teams at Wyss Institute at Harvard University and Korea Institute of Science and Technology[5][6].
