Dorzagliatin is a novel, first-in-class, dual-acting allosteric glucokinase activator (GKA) developed for the treatment of diabetes. It works by directly binding to a site on glucokinase (GK) that is distal to the active site, increasing GK's affinity for glucose and enhancing its glucose-sensing capacity. This action restores impaired glucose homeostasis by improving both pancreatic β-cell function and hepatic glucose metabolism in a glucose-dependent manner. Dorzagliatin has demonstrated efficacy in reducing HbA1c and improving insulin secretion in patients with type 2 diabetes mellitus (T2DM), and it has also shown potential benefits for type 1 diabetes mellitus (T1DM) and glucokinase-maturity-onset diabetes of the young (GCK-MODY). The drug was developed by Hua Medicine and is approved in China as monotherapy or as an add-on to metformin for glycemic control in adults with T2DM[1][2][3][5][6].
