Dorzagliatin + small molecule GLP-1 is a combination therapy under development by Hua Medicine for the treatment of type 2 diabetes and obesity. The combination consists of dorzagliatin (HMS5552), a first-in-class glucokinase activator (GKA), and a proprietary small molecule glucagon-like peptide-1 receptor agonist (GLP-1RA), such as HMS-1649. Dorzagliatin acts as a glucose sensor restorer by activating glucokinase in the pancreas, liver, and intestine, thereby improving insulin secretion and glucose metabolism. The small molecule GLP-1RA component enhances glucose-dependent insulin secretion, inhibits glucagon release, and slows gastric emptying, while also providing weight loss benefits through appetite suppression. This oral combination therapy is designed to provide a synergistic approach to metabolic control by targeting multiple physiological pathways involved in glucose homeostasis and energy balance.
