Overview

Dostarlimab + encelimab is an investigational combination of two monoclonal antibodies for the treatment of advanced or metastatic solid tumors. Dostarlimab is a humanized IgG4 monoclonal antibody that targets and inhibits programmed death receptor-1 (PD-1), thereby blocking its interaction with PD-L1 and PD-L2 to enhance T-cell mediated anti-tumor immune responses. Encelimab (TSR-033) is a novel anti-LAG-3 monoclonal antibody that inhibits lymphocyte activation gene 3 protein (LAG-3), another immune checkpoint involved in downregulating T-cell activity. The rationale for combining these agents is to achieve dual checkpoint blockade—targeting both PD-(L)1 and LAG-3 pathways—to potentially improve anti-tumor efficacy in patients with advanced cancers who have progressed on prior therapies. Clinical studies have shown manageable safety profiles but limited clinical activity in heavily pretreated populations[1][2][4].
